The compound reliably triggers a short pulse of growth hormone, but almost everything people want from that pulse, such as fat loss, muscle, better sleep and faster recovery, rests on inference and anecdote rather than on trials of ipamorelin itself. Notice what is missing from the top of the table. No benefit that people actually buy ipamorelin for has a controlled human trial behind it. A 2026 narrative review of the peptides used for GH-IGF-1 modulation made the same point: it ranks them by evidence tier, from regulatory-grade trial data to no human studies at all, and stresses the uncertainty around performance and recomposition benefits (PMID 42395176). Our own search of Europe PMC for ipamorelin with sleep, body composition or IGF-1 and human data turned up reviews, not trials. That search was not exhaustive, so read it as "we did not find one".

What the human data actually show

Two human studies carry the weight. The first, a 1999 dose-escalation study, gave five intravenous infusion rates over 15 minutes to eight healthy men each. Converting the published figures, that is roughly 3 to 100 mcg/kg. Ipamorelin released growth hormone at every level, in a single episode peaking around 40 minutes after the start, with a terminal half-life of 2 hours (PMID 10496658). That is solid evidence of a hormone effect. It is also a narrow one: it measured GH, not IGF-1, body composition or sleep.

The second is a 2014 phase 2 trial that tested ipamorelin for postoperative ileus, a stalled gut after surgery. Patients received 0.03 mg/kg intravenously twice daily for up to seven days. The drug was well tolerated, and the primary endpoint, time to tolerating a solid meal, was not significantly different from placebo (25.3 versus 32.6 hours, p = 0.15). The authors noted the study was small and mixed several conditions (PMID 25331030). That result matters for benefits in two ways. It shows ghrelin-receptor stimulation did not translate into a measurable clinical gain in that setting, and it reminds you that a plausible mechanism and a proven benefit are different things.

Animal-only findings

In swine, ipamorelin released growth hormone without raising ACTH or cortisol, even at more than 200 times the dose needed for the GH effect, while two older peptides in the class did raise them. That is the basis of the "selective" label (PMID 9849822). It has not been shown in people, and the Reddit data are not a substitute: several commenters in this sample describe flushing, anxiety or a racing heart, which they cannot attribute to a specific mechanism.

The bone findings are also from rats. Adult female rats given ipamorelin for 15 days had dose-dependent increases in tibial growth rate, from 42 micrometres a day on vehicle to 52 on the top dose, and the total IGF-I level did not change (PMID 10373343). In a 12-week study, ipamorelin and GHRP-6 raised bone mineral content, but the increase came from larger bones, not denser ones (PMID 10828840). A third study found that ipamorelin offset some of the bone and muscle loss caused by a steroid in rats (PMID 11735244). These are interesting research directions. None shows that a person injecting ipamorelin gains bone.

The IGF-1 line deserves a note. Many guides say ipamorelin raises IGF-1 with moderate evidence. The human study we could verify measured GH, and the rat study found no change in total IGF-I, so we put IGF-1 at the anecdote tier until a controlled human measurement turns up.

Claimed but unproven: fat loss, muscle, sleep and anti-aging

The usual argument runs: ipamorelin raises GH, GH supports fat breakdown and tissue repair, so ipamorelin should help body composition. Each step is plausible, but the conclusion has not been tested for ipamorelin alone in people we could find. Pulsatile GH from a secretagogue is also capped by what the pituitary holds, which is part of why users and researchers expect a modest effect compared with injected growth hormone.

Sleep has a similar story. GH release is tied to deep sleep, so a bedtime dose seems like it should help. Nothing we verified measured that. The Reddit reports below are the only data, and they point both ways.

Anti-aging and "longevity" claims have the weakest footing of all. We found no human data on ipamorelin and ageing outcomes, and the long-term effects of repeated GH-axis stimulation are unknown. If a source gives you a percentage improvement from ipamorelin alone, ask where the trial is.

What Reddit users say they notice

To see what people describe, we read 456 posts and comments from 24 threads in the benefits search and 302 more from the dosing search, across 14 subreddits in total. Most were noise: stacking cheat sheets, a long-COVID thread, vendor posts and questions. Thirty comments from 9 threads and 6 subreddits (mainly r/BodyHackGuide) described first-hand what happened on ipamorelin, almost always combined with CJC-1295. Only 7 of the 30 came from the benefits search; the rest are from dosing threads that included outcomes. We excluded a very detailed "my data" post that claimed a 40 percent rise in deep sleep, because its numbers were unverifiable and its wording read like promotion.

Sleep was the most common report in both directions. One commenter on a blend wrote that there was "no improvement in sleep, energy, no increase in fat loss, mental clarity, etc." after taking 200 mcg five nights a week (a r/Peptidesource commenter, Dec 2025, link). That commenter did describe a warm facial flush and light-headedness for ten minutes after each injection. Another commenter said they noticed no change after about two months. On the other side, ten accounts reported better sleep, among them people who noticed it within the first week.

Body composition reports were small and confounded. Two of the five accounts describing a leaner or fuller look were also on tirzepatide, which would explain much of it, and one commenter who tracked scans over four weeks said they dropped 1 percent body fat and gained a pound of muscle while training six days a week. Five accounts described less soreness or better workout energy, and two described sluggish cardio early on. Three mentioned puffiness or fluid retention.

Lab reports are rare and mixed. A r/NTNPerformance commenter (May 2026) wrote: "my IGF definitely increased, not as much as I wanted to, so now I'm just going to try HGH straight." (link). A second user said IGF-1 went from normal to near the top of the scale on 100 mcg each of ipamorelin and CJC-1295 at night. Two lab reports cannot be generalised, but they are among the few objective numbers in the sample.

Negative reports also appeared. Nine accounts described anxiety, broken sleep, a racing heart or being awake at 2 am, usually after raising a dose or injecting at a bad time, and one person reported an allergic reaction after about eight weeks. Why two people report different results

Reported benefits depend on more than the molecule. Almost everyone combined ipamorelin with CJC-1295, so the effects cannot be assigned to one peptide. The honest answer is that nobody has measured it. The accounts below are single data points, and they range from a first-week change to nothing at two months. One commenter described feeling poor for the first two to four weeks before things turned: "After I started week 5 it was a drastic change." (a r/BodyHackGuide commenter, Mar 2026, link).

Several people advised waiting a month before judging. That advice may be sound, but a slow start is also exactly what a placebo response, a change in training, or no effect at all would look like. Without a control arm, none of these timelines can distinguish them.

Honest limits

Four limits apply to everything above. First, the human GH data come from a handful of intravenous studies, none of which reproduces the subcutaneous routine people follow. Second, no controlled study of ipamorelin on fat, muscle, sleep or recovery turned up in our search. Third, long-term human safety is unstudied, and growth hormone stimulation can affect fluid balance and insulin sensitivity, so anyone with diabetes, a cancer history or a prescription medication list should speak to a clinician. Fourth, every Reddit number here comes from an unverified, self-selected group of people who chose to post, so the percentages describe who wrote, not what is likely to happen to you.
